Biscuits or just carpenter glue for arcade stick box?

Finally ran out of Red Octanes so it’s time to build some from scratch. The link is similar to what I’m doing except my cuts will be 90 degrees and I will be using the little blocks to support the top panel and bottom cover. Will carpenter glue and some clamps over night be strong enough? And do I glue the top panel for the controls and and glue the support blocks with the box or wait until it dries? Should I put clamps on the support blocks while the glue dries or do you just set them where you want them?

you can glue, but I prefer to use dowels.

Or if you have a brad nailer, you can use that in addition to glue if you are painting.
